Understanding Aunt and Uncle DNA Testing
An aunt or uncle shares DNA with a biological niece or nephew because both descend from the same family line. A full sibling of the alleged father inherited DNA from the same two parents as he did, although not in exactly the same combination.
The laboratory examines the amount and pattern of DNA shared between the tested child and possible aunt or uncle. It then calculates whether those findings are more consistent with an avuncular relationship or with the participants being unrelated.
What the test can and cannot establish
An avuncular test evaluates the specific relationship described in the case. It does not literally recreate the missing father’s complete DNA profile, and it cannot promise the same statistical power as a direct paternity test.
The laboratory must know whether the tested aunt or uncle is:

Improving the result
Please include the child’s mother whenever possible as this will greatly improve the statistical data. Mother is normally needed to consent for a child under the age of 16 years unless another person with proved Parental Responsibility is available.

Accuracy and reliability
The reliability of an avuncular test comes from correct participant identities, an accurate family history, good sample collection, suitable genetic markers and appropriate statistical interpretation.
Be cautious of providers advertising a guaranteed inclusion percentage before seeing the participants’ DNA. Complex relationship cases do not all produce the same amount of information and a direct paternity DNA test would be the most conclusive option if the alleged father is available for testing.
At-home and legal testing
For personal knowledge testing, we are happy to send a sampling kit directly to the participants for them to take cheek swabs at home. Each adult must consent, and somebody with Parental Responsibility must provide the required permission for a child, evidence of Parental Responsibility will be required if the child's mother is unavailable.
For court, immigration or another official purpose, self-collection is not acceptable. Please let us know if a legally approved report is needed so a witnessed, chain-of-custody case can be arranged where available.
